Dolutegravir, emtricitabine and tenofovir alafenamide is a three-drug combination of dolutegravir (integrase strand transfer inhibitor [INSTI]), emtricitabine (EMT) and tenofovir alafenamide (TAF), these two are HIV nucleoside analog reverse transcriptase inhibitors (NRTIs) and is indicated for use alone as a complete regimen for the treatment of HIV-1 infection in adults and pediatric patients. Dolutegravir-based treatment is being introduced as the preferred first-line regimen in many countries as per World Health Organization recommendations. Dolutegravir is preferred as the basis of first-line treatment because of high effectiveness, a high barrier to resistance and few side effects. This project proposal aims to develop and validate a LC- MS/MS method for the determination of EFV, EMT, TDF, DTG and TAF in human plasma samples from HIV patients and to apply the method in real samples from Botswana for therapeutic drug monitoring and pharmacokitic profiling. Human blood plasma will be extracted, and chromatography will be performed on a C18 column using a suitable gradient program and mobile phase for separation.
